    How do you do the "laser effect", like in this video at 0'43 ?

    If I wanted to do this, I'd drop a graphic of the beam on a track above and then use masking to reveal it at the appropriate time.

    As Andy says, but you could use the chroma key.

    But it would be much more fun to use a real laser

    Strictly speaking, of course, that effect is nothing like a laser. One of the key aspects, if not the defining aspect, of a laser is that it is a beam of energy which does not get wider as that in the video does.
